Title: Atividade do óleo de copaíba no modelo de asma alérgica experimental induzido por ovalbumina em camundongos BALB/c

Resumo: A asma é uma doença inflamatória alérgica das vias aéreas, considerada um problema de saúde pública, capaz de causar sofrimento significativo e altos custos para as famílias e para os sistemas de saúde. Problemas relacionados à dificuldade de acesso ao tratamento, além de baixa adesão e risco de baixa responsividade, em certos casos, justificam a pesquisa de novos compostos com propriedades anti-inflamatórias e imunomodulatórias, mais acessíveis e de baixo custo. O óleo de copaíba (OC) é uma alternativa renovável, de origem natural, já amplamente utilizada na medicina popular com atividade anti-inflamatória, mas ainda não estudada para o tratamento desta doença. Através do modelo de asma alérgica com ovalbumina (OVA), que permite a indução das principais respostas das células T auxiliares (TH) envolvidas nesta inflamação, investigamos os efeitos do OC em camundongos BALB/c. O óleo essencial investigado teve sua composição verificada por cromatografia gasosa com detector por ionização em chama (CG-FID) e cromatografia gasosa acoplada ao espectrômetro de massas (CG-MS). Testes para toxicidade hepática, renal ou efeitos glicêmicos foram realizados nos camundongos empregando-se o OC por gavagem nas dosagens a serem utilizadas na investigação, 50 mg/kg e 100 mg/kg. Após a indução da asma alérgica, o potencial anti-inflamatório do OC foi avaliado pela contagem total e diferencial das células inflamatórias no lavado traqueal, pela produção de óxido nítrico (NO) e pela concentração das citocinas relacionadas a patogênese da asma alérgica, interleucina-4 (IL-4), interleucina-17 (IL-17), interferon-γ (IFN-γ) e fator de necrose tumoral-α (TNF-α), no lavado traqueal e no macerado pulmonar. Análises histopatológicas e histomorfométricas também foram realizadas em amostras pulmonares. A análise dos resultados obtidos mostrou que o tratamento com OC atenuou significativamente os sinais inflamatórios, reduzindo a contagem de células inflamatórias presentes no lavado traqueal, suprimindo a produção de NO e reduzindo a resposta mediada por células T, envolvidas na doença. Nos animais tratados houve também redução do infiltrado inflamatório no tecido pulmonar, com redução da área de parênquima alveolar. Não foi possível, entretanto, correlacionar um efeito dose-reposta devido ao fato de não serem demonstradas diferenças estatísticas entre as duas doses utilizadas (50 e 100 mg/Kg) na maioria dos resultados. Com base no exposto, este estudo concluiu ser o OC uma substância promissora no tratamento da asma, sendo ainda necessários estudos mais aprofundados para a elucidação completa do seu efeito no organismo.
Abstract: Asthma is an inflammatory allergic disease of the airways, considered a public health problem, capable of causing significant suffering and high costs for families and for health systems. Problems related to the difficulty of access to treatment besides low adherence and risk of low responsiveness in certain cases, justify the research of new compounds with anti-inflammatory and immunomodulatory properties, including accessible and low cost. Copaiba oil (OC) is a renewable alternative of natural origin widely used in folk medicine with anti-inflammatory activity but not yet studied for the treatment of this disease. Through the model of allergic asthma with ovalbumin (OVA), which allows the induction of the main T helper cells (TH) responses involved in this inflammation, we investigated the effects of OC in BALB/c mice. The essential oil investigated had its composition verified by gas chromatography with flame ionization detector (CG-FID) and gas chromatography coupled to the mass spectrometer (CG-MS). Tests for hepatic, renal, or glycemic effects toxicity were performed in the mice using OC by gavage at the dosages to be used in the investigation, 50 mg/kg and 100 mg/kg. After the induction of allergic asthma, the anti-inflammatory potential of OC was evaluated by the total and differential count of the inflammatory cells in the tracheal lavage, nitric oxide (NO) production and concentration of cytokines related to the pathogenesis of allergic asthma, interleukin-4 (IL-4), IL-17, interferon-γ (IFN-γ), and tumor necrosis factor-α (TNF-α) in the tracheal lavage and pulmonary maceration. Histopathological and histomorphometric analysis were also performed on pulmonary samples. The analysis of the results showed that the OC treatment significantly attenuated the inflammatory signals, reducing the count of inflammatory cells present in the tracheal lavage, suppressing NO production and reducing the T cell-mediated response involved in the disease. In the treated animals there was also a reduction of the inflammatory infiltrate in the lung tissue, with reduction of the alveolar parenchyma area. It was not possible, however, to correlate a dose-response effect due to the fact that statistical differences between the two doses (50 and 100 mg/kg) were not demonstrated in most of the results. Based on the above, this study concluded that OC is a promising substance in the treatment of asthma, and further studies are needed to fully elucidate its effect on the body.
