Resumo: A adição sulfa-Michael consiste na adição de um nucleófilo de enxofre a uma ligação múltipla ativada por um grupo retirador de elétrons, levando a formação da ligação C-S. Várias abordagens catalílicas foram desenvolvidas usando tióis como nucleófilos. Especificamente no caso da metilsulfenilação, o metanotiol ou o seu sal de sódio são os nucleófilos mais apropriados para essa reação. De forma alternativa, sulfeto de hidrogênio pode ser usado se seguido de metilação na presença de uma base. Embora estejam bem estabelecidos, esses reagentes são caros, difíceis de manusear e apresentam alta toxicidade e baixo ponto de ebulição. Vários trabalhos na literatura apontam o dimetilsulfóxido (DMSO) como um reagente de metilsulfenilação bastante promissor por ser um solvente comercialmente disponível, de fácil manuseio e significativamente mais barato que outros reagentes de enxofre. Entretanto, não há relatos sobre a aplicação deste em reações de sulfa- Michael. Nesse trabalho são apresentadas três novas metodologias de metilsulfenilação em sistemas α,β-conjugados e carbono saturado, empregando DMSO como fonte de enxofre. A primeira metodologia foi desenvolvida em meio às tentativas de triclorometilação de dibenzilideno acetonas (DBA’s). Pela análise de espectros de RMN, espectrometria de massas de alta resolução (EMAR) e experimentos controle, concluiu-se que o produto formado era metilsulfenilado. Com a finalidade de avaliar a generalidade dessa reação, substratos como diferentes chalconas, DBA’s, derivados de aduto de Morita-Baylis-Hillman (MBH) foram empregados em presença de sal de tricloroacetato de portássio (KTCA), ácido canforsulfônico (ACS) como organocatalisador, ácido acético a 100 °C, peneira molecular de 4Å e DMSO como solvente e reagente de metilsulfenilação, sob irradiação micro-ondas. Na tentativa de elucidar o mecanismo, estudos experimentais e cálculos teóricos empregando o método DFT foram realizados. A partir dos resultados destes estudos foi possível propor um mecanismo que passa por uma deoxigenação do DMSO e formação do íon sulfênio, espécie chave nesse trabalho. Este íon foi interceptado quando substituiu o ácido acético por um ácido graxo que levou a formação do éster de metil tiometila (MTM). A segunda metodologia de metilsulfenilação apresentada nesse trabalho consiste em uma reação empregando chalconas, éster de MTM, ACS, acetato de sódio, ácido acético a 100 ° C, peneira molecular de 4Å e DMSO. O éster de MTM é necessário, porém o seu papel não é bem compreendido. A terceira metodologia é mais simples e envolve derivados de adutos de MBH. Nessa reação é utilizado apenas ácido acético, acetato de sódio e DMSO. Todas as metodologias são eficientes, inéditas e levam a formação de produtos sulfenilados em rendimentos de moderados a elevados.
Abstract: The Sulfa-Michael addition consists of a sulfur nucleophile addition to an activated multiple bond by an electron withdrawing group, leading to the formation of the C-S bond. Several catalytic approaches have been developed using thiols as nucleophiles. Specifically, in the case of methylsulphenylation, methanethiol or its sodium salt would be the most appropriate nucleophiles for this reaction. Alternatively, hydrogen sulfide may be used followed by methylation in the presence of a base. Although well established, these reagents are expensive, difficult to handle, and exhibit high toxicity and low boiling point. Several papers in the literature point out the utility of Dimethyl sulfoxide (DMSO) as a promising methylsulphenylation reagent because it is a commercially available, easy to handle and significantly cheaper than other sulfur-based reagents. However, there are no reports of its application to sulfa-Michael reactions. In this work, three new methylsulphenylation methodologies using α,β-conjugated systems and saturated carbon, DMSO as the source of sulfur are presented. The first methodology was developed during trichloromethylation attempts of dibenzylidene acetones (DBA's). By analysis of NMR spectrum, high-resolution mass spectrometry (HRMS) and control experiments, it was found that the product formed was methylsulfenylated derivative. In order to investigate the scope of this reaction, substrates such as different types of chalcones, DBA's, MBH adduct derivatives were employed in the presence of the salt of trichloroacetate (KTCA salt), camphorsulfonic acid (CSA) as organocatalyst, acetic acid at 100 °C and DMSO as solvent and methylsulphenylation reagent, under microwave irradiation. In an attempt to elucidate the mechanism, experimental studies and theoretical calculations using the DFT method were performed. Through these results it was possible to propose a mechanism that passes for a deoxygenation of DMSO and formation of the sulfonium ion, key species in this work. This ion was intercepted when the acetic acid was replaced by a fatty acid which led to the formation of the MTM ester. The second methylsulphenylation methodology consists of a reaction employing chalcones, MTM ester, ACS, sodium acetate, acetic acid at 100 °C and DMSO. The MTM ester is necessary, but its role is not well understood. The third methodology is simpler and involves derivatives of MBH adducts. In this reaction only acetic acid, sodium acetate and DMSO are required. All methodologies are original, efficient and lead to the formation of sulfenylated products in moderate to high yields.
